Given that my Endurance is already at 43 to help cover the equipment that I choose to lug around, would equipping the Ring of Favor and Protection increase my stamina passed the 40 cap, or as I am beginning to suspect would the only benefit be increased HP along with increased Equipment Load?

RoFaP will increase your stamina past the normal stamina cap, up to 192.

And you know this for sure, as in you've seen this in practice, and you're not just going by what the DkS Character Planner says?

Yes it definitely does work like that.
You can even see it when you highlight the RoFaP without equipping it. It'll show that your stamina will go up to 192.
